【摘要】 通过正交试验确定了白背毛木耳(Auricularia polytricha)43-26液体发酵培养基的最优配方(g/L):土豆20、玉米粉20、蛋白胨10和磷酸二氢钾5;利用超声波辅助水提醇沉法提取胞内粗多糖,多糖含量为75.5%;多糖粗提物经DEAE-52纤维素离子交换层析和Sephadex G-200层析分离纯化,分别得到4个相对均一的纯多糖组分AP1、AP2、AP3和AP4。
【Abstract】 A growth medium for maximizing polysaccharide production by submerged cultures of Auricularia polytricha 43-26,consisting of(g/L) 20.0 potato,20.0 corn powder,10.0 peptone and 5.0 KH2PO4,was derived by orthogonal testing.A crude fraction containing 75.5% polysaccharide was obtained by aqueous extraction of ultrasound-treated fungal mycelium and precipitation with ethanol.Crude polysaccharide was separated into four partially purified fractions,AP1,AP2,AP3 and AP4,by DEAE-cellulose 52 and Sephade...
