【中文摘要】 以温度40℃处理为实验组,10℃处理为对照组,采用双向电泳技术,比较了虎奶菇菌丝体蛋白质的3种提取方法,其中TCA/丙酮法提取所得到的蛋白质样品的双向电泳图谱效果最佳,为其它大型真菌蛋白质的提取提供了重要参考。并对TCA/丙酮法加以改进,获得虎奶菇菌丝体胞外蛋白质样品的制备方法,为应用差异蛋白质组学方法对虎奶菇菌丝体自溶进行研究奠定基础。
【英文摘要】 In this study,the Pleurotus tuber-regium(Fr.) Sing mycelia processing 40℃was used as test group and those with 10℃treatment served as conrol group.Three methods of extracting mycelium proteins were compared.The results showed that the TCA/acetone method is optimal with regard to the electrophoretic effect of protein sample,which provided useful guidance for the protein prepartion of other macromycetes.The TCA/acetone method was adjusted and improved in this study to be suited for extracting the extracellular proteins of the Pleurotus tuber-regium(Fr.) Sing mycelium,providing the basis for investigating the mycelium autolysis of the Pleurotus tuber-regium(Fr.) Sing with differential proteomic method.
