    温度胁迫对白灵侧耳菌丝保护酶活性的影响

    【摘要】 以白灵侧耳(Pleurotus nebrodensis)菌株中农一号(CAAS 1)为试验材料,研究了温度胁迫(4℃和35℃)对菌丝体内超氧化物歧化酶(superoxide dismutase,SOD)、过氧化物酶(peroxidase,POD)和过氧化氢酶(catalase,CAT)活性的影响。结果表明,温度胁迫导致3种保护酶活性均发生显著的变化。SOD活性随温度胁迫时间的延长先急剧升高后下降,在胁迫12 h时均达到最大值(低/高温胁迫分别为46.23 U/g和50.33 U/g);POD活性呈现先升高后缓慢下降的趋势,在胁迫处理24 h时达到最高值(低/高温胁迫分别为6.86 U/g和7.22 U/g);CAT活性在胁迫处理12 h内迅速上升,之后平稳上升。

    【Abstract】 Superoxide dismutase(SOD),catalase(CAT) and peroxidase(POD) activities were determined over 60 h time courses in extracts of Pleurotus nebrodensis(strain CAAS 1) mycelia incubated at 4 ℃,25 ℃(controls) or 35 ℃ following growth at 25 ℃ for 14 d.SOD activities in extracts of mycelia incubated at 4 ℃ and 35 ℃ increased markedly during the first 12 h following temperature shift and declined thereafter,whereas very little fluctuation in enzyme activity was observed in extracts of mycelium continually maintained at 25 ℃.POD activity increased in extracts of mycelia incubated at all three temperatures and reached peak values after 24 h before declining throughout the next 24 h in mycelium incubated at 35 ℃ and throughout the next 36 h in mycelia incubated at both 4 ℃ and 25 ℃.CAT activity in extracts of mycelia incubated at 4 ℃and 35 ℃ also increased markedly during the first 12 h following temperature shift and continued to slowly increase in the latter case.However,enzyme activity in extrac...更多ts of mycelium continually maintained at 25 ℃ remained at much lower,relatively constant,levels throughout.CAT and POD activities in mycelia incubated at 4 ℃ and 35 ℃ were significantly(P <0.01) higher than values recorded in mycelium maintained at the original growth temperature(25 ℃) throughout the 60 h monitoring period.SOD activities were significantly(P <0.01) higher for 48 h and 36 h in extracts of mycelia incubated at 35 ℃ and 4 ℃,respectively compared with values recorded in mycelium maintained at the original growth temperature(25 ℃).Our data show that temperature shifts to higher and lower values have a significant effect on the activities of antioxidative enzymes in P.nebrodensis mycelia.  还原
